Protein conformation is important to its function, for therapeutic proteins the three-dimensional (3-D) structure is closely related to their safety and efficacy. Considerable resources are invested in the evaluation of therapeutic protein immunogenicity, a major concern in biologics development. Many technologies have been used to characterize therapeutic protein 3-D structure, the most commonly used technologies include Near UV CD, Analytical Ultracentrifugation, Gel Filtration (Size Exclusion), NMR and Hydrogen/deuterium Exchange (HDX). However a more sensitive, systematic and high throughput method will be beneficial to the development of large number of biologics molecules or to characterize each biologics in much detail.
